Cell Culture Media

Cell culture media is the nutrient-rich liquid environment that sustains living cells outside the body. It provides everything a cell needs to survive and proliferate: glucose for energy, amino acids for protein synthesis, vitamins, salts, and trace minerals for metabolic function. Most formulations also include a buffering system, typically sodium bicarbonate in combination with 5% CO₂, to maintain the slightly alkaline pH (7.2–7.4) that mammalian cells require.

Beyond the basal nutrients, most cell culture media are supplemented with serum: most commonly fetal bovine serum (FBS), which contributes a complex cocktail of growth factors, hormones, and carrier proteins that promote cell attachment and proliferation. The exact formulation varies widely depending on the cell type: HeLa cells, CHO cells, and primary neurons, for example, each have distinct nutritional demands. Common basal media formulations like DMEM, RPMI-1640, and MEM have been refined over decades to meet those specific needs.

Maintaining media quality is critical to experimental reproducibility. Contamination by bacteria, fungi, or mycoplasma can alter cell behavior and invalidate results, which is why sterile technique,sterile filtering, and routine testing are standard practice. Media components also degrade over time. Glutamine breaks down into ammonia, which is toxic to cells, so freshness and proper storage matter as much as the formulation itself. In biopharmaceutical manufacturing, where cells are used to produce therapeutic proteins and antibodies, media optimization is a major lever for maximizing yield and product quality.
